The antiviral activities of recombinant human interferon-¥á (rHu/IFN-¥á) were assayed on the various cells that were infected with six different viruses (vesicular stomatitis virus : VSV, herpes simples virus type I : HSV-I, encephalomyocarditis virus D strain : EMC-D, human rotaviruses : Wa and DS-1, mouse hepatitis virus : MHV). Also antitumor effects of rHu/IFN-¥á on established human tumor cell lines(KU812, K 562, RPMI and MOLT : human T-cell leukemia, AU72 : human malignant melanoma) were investigated by measuring in vitro growth inhibition of the cells after rHu/IFN-¥á treatment.
Potent antiviral activity was demonstrated on Vero and HEp-2 cells infected with VSV and HSV-1. No or very low antiviral activity was demonstrated on L-929 cells infected with VSV, HSV-1 and EMC-D. Also potent antiviral activity was demonstrated on MA104 and Vero cells infected with human rotaviruses.
The sensitive tumor cell lines against rHu/IFN-¥á were RPMI and AU72 since 10©ø¡10^5IU/ml produced 62.3¡76.0% growth inhibition of the cells after 24 hours with rHu/IFN-¥á treatment. On the other hand, after 24 hours with rHu/IFN-¥á treatment, there were no or very low growth inhibition effects of KU812, K562 and MOLT tumor cell lines at the concentration of 10©ø¡10^5IU/ml of rHu/IFN-¥á.
